Although gaining much attention in recent years, it is unclear whether mycorrhizal fungi distribute meaningful amounts of resources among trees in ways that increase the fitness of the receiving trees. To this end, we used shaded and non-shaded pairs of inter- and intra-species Pinus halepensis and Quercus calliprinos saplings growing outdoors in forest soil. Carbon transfer was measured using pulse labeling with 13CO2 and the mycorrhizal community of each tree was identified by DNA barcoding. The effects of belowground connections were examined by tree performance and Non-Structural Carbohydrates (NSC) pools. Although we did not observe any growth benefits, shaded recipient oaks exhibited higher levels of root NSC compared to their control counterparts, which were not connected belowground. This finding suggests a potential benefit of establishing below-ground connections. We also show that non-shaded pines connected to shaded oaks were depleted of their starch pools, suggesting a possible cost of the tree-fungi-tree interaction. Additionally, we monitored the carbon (C) flow from a 13CO2 labeled donor pine tree to the final recipient oak tree and were able to demonstrate C transfer from pines to shaded oaks. Finally, we were able to identify the main fungal symbionts interacting with pines and oaks. Our results link specific mycorrhizal species to belowground C transfer and suggest C-driven fitness costs and benefits to the trees.

Background: As the population of people with CF (pwCF) continues to age, attention is shifting towards addressing the unique challenges teenagers and adults face, including substance use. Changing attitudes and legality regarding marijuana and CBD may influence their use among pwCF, but data on their prevalence, reasons for use, and administration methods are lacking. Objective: Investigate marijuana, cannabidiol (CBD), e-cigarette, and cigarette usage among pwCF and explore differences in demographics, disease severity, and CFTR modulator use between current and non-users. Methods: This cross-sectional study used a one-time electronic survey to assess marijuana, CBD, e-cigarette, and cigarette use in pwCF aged >13 years. Demographic and clinical characteristics were compared between current users and non-users. The association between current substance use and CFTR modulator use was analyzed using logistic regressions. Results: Among 226 participants, 29% used marijuana, 22% used CBD, 27% used e-cigarettes, and 22% used cigarettes. Current users of all substances were more likely to be college-educated, Black, or aged 29-39 years than non-users. Current e-cigarette users were 2.9 times more likely to use CFTR modulators (95% CI 0.98-11.00, p=0.08) and current marijuana users were 2.5 times more likely to use CFTR modulators compared to non-users, adjusted for confounders. Current users of CBD, e-cigarettes, and cigarettes were more likely to have an abnormal mental health screen compared to non-users. A high proportion of never-users of marijuana and CBD expressed interest in using. Conclusion: Substance use is more prevalent among pwCF than previously reported and needs to be addressed by healthcare providers.

Background Adolescents living with obesity (AlwO) can have limited exercise capacity. Exercise capacity can be predicted by a 2-factor model comprising lung function and leg muscle function, but no study has looked at cycling leg muscle function and its contribution to cycling exercise capacity in AlwO. Methods 22 nonobese adolescents and 22 AlwO (BMI>95 percentile) were studied. Anthropometry, body composition (DEXA), spirometry, 30-sec isokinetic work capacity, and maximal exercise (cycle ergometry) were measured. Results AlwO had greater lean leg mass (LLM) (14.8±4.1 vs 21.0±4.3 kg, Con vs AlwO p<0.001). Lung function did not differ, although FEV 1 trended higher in AlwO (101.0±13.1 vs 107.9±12.7 percent predicted, p=0.08). Leg 30-second work output did not differ in absolute terms or per allometrically scaled LLM. Peak oxygen consumption did not differ between the groups in absolute terms or as percent predicted values (78.5±15.4 vs 82.1±16.5 percent predicted), but was lower in AlwO when expressed per scaled kg of LLM. Peak oxygen consumption related to both lung function and 30-second work capacity, with no observed group effect. 30-second leg work capacity related to the scaled muscle mass, with a small group effect. There was some correlation between leg work capacity and time spent in moderate to vigorous physical activity in AlwO (r s=0.39, p=0.07). Conclusion AlwO have larger LLM and preserved exercise capacity, when expressed as percentage of predicted, but not per allometrically scaled LLM. Increasing time spent in moderate to vigorous activity may benefit AlwO.

African science has substantial potential, yet it grapples with significant challenges. Here we describe the establishment of the Biomedical Science Research Centre (BioRTC) in Yobe State, Northeast Nigeria, as a case study of a hub fostering on-continent research and describe strategies to surmount some of these barriers. We detail the steps taken to establish BioRTC, emphasising the critical importance of stakeholder engagement, community involvement, resource optimisation, and collaborations. Although we are in the early stages of our journey, our experience can nonetheless serve as a guide to others embarking on similar endeavours in resource-limited settings. We invite the support and collaboration of those who share our vision and believe in our potential.

Acute thrombus formation on the delivery sheath is rare condition during percutaneous left atrial appendage occlusion. We presented two cases that TEE showed a floating thrombus attached to the tip of delivery sheath during the procedure. Cerebral embolic protection devices were used to prevent neurological events after thrombus was detected. The

Reservoir is a vital tool for human utilization of water resources, and the deterioration of its water quality can seriously threaten the water cycle and sustainable urban development. However, there are relatively few studies in academia that analyze and evaluate the water quality of multiple reservoirs at the same time. To address this knowledge deficit, we collected 108 water samples from three different reservoirs in Chuzhou City for 36 months (from 2019 to 2021), explored the drivers of changes in water quality parameters over time and the extent of eutrophication. Our results indicated that the water quality of the reservoirs was deteriorating during the study period, among which Huanglishu Reservoir and Shahe Reservoir reached mild eutrophic status, and both had higher eutrophication levels than Chengxi Reservoir. Secchi Depth (SD), Total Nitrogen (TN), and Total Phosphorus (TP) were the principal factors inducing eutrophication. The biased utilization of reservoir functions was the major contributor to the discrepancy in the degree of eutrophication. Furthermore, Pearson Correlation Analysis revealed that there are significant correlations between many water quality parameters. Cluster Analysis (CA) grouped the 12 months of each year into three clusters (stable water level period, rainy season high flow period, and winter low flow period). Based on this, Analysis of Variance (ANOVA) showed that most water quality parameters varied considerably between the clusters. Collectively, this study identified the actual water quality conditions of three reservoirs in Chuzhou City and provided guidance for local water quality management and environmental protection.

TENOFOVIR INDUCED FANCONI SYNDROME IN A MIDDLE AGE AFRICAN FEMALE FROM KENYA, EAST AFRICA: CASE REPORT AND BRIEF LITERATURE REVIEW.

Objectives: Congenital ventricular outpouching (CVO) is a rare cardiac malformation that can manifest as congenital ventricular aneurysm (CVA) and/or congenital ventricular diverticula (CVD). In this study, we describe the prenatal features and postnatal follow-up of 27 cases of CVO. Methods: The clinical data of 27 patients with CVO who attended Sir Run Run Shaw Hospital Affiliated to the Medical College of Zhejiang University (Zhejiang Province, China) and Taizhou Hospital of Zhejiang Province Affiliated to Wenzhou Medical University (Zhejiang Province, China) from April 2013 to October 2022 were retrospectively analyzed. The patients were also followed up by telephone. The prenatal characteristics and postnatal outcomes of the patients with CVO were evaluated. Results: CVO was detected in 26 cases prenatally,, 14 (51.85%) were diagnosed with CVA, 9 (33.33%) were diagnosed with CVD, 3 (11.11%) were equivocal for CVA/CVD, and 1 (3.70%) was detected with CVA postnatally. Six patients underwent follow-up fetal echocardiography approximately 4 weeks after the initial echocardiography examination, and a significant difference in CVO size was observed between the two examinations ( P = 0.02). Eight patients (29.63%) demonstrated cardiovascular dysfunction, and the median CVO size in fetuses with and without cardiovascular dysfunction was 205 (range: 169–396) mm 2 and 124 (range: 92–154.5) mm 2, respectively ( P = 0.01). Eight patients (29.63%) had cardiac/extracardiac defects. Thirteen patients were live born, 12 were terminated pregnancies, and 2 were lost to follow-up. The postpartum size of the CVOs remained stable in six patients, decreased in two patients, dissolved in three patients, and were surgically removed in two patients. With the exception of one patient with CVA complicated with complex congenital cardiac malformation who underwent surgical treatment after birth and who had postoperative left ventricular dysfunction (Case 1), the prognosis of all of the patients was good. Conclusion: Most cases of fetal CVO have typical echocardiographic manifestations of CVA or CVD, while some cases manifest as a mixture of the two. The size of fetal CVO can increase with the increase in gestational age. The occurrence of cardiovascular dysfunction is related to the CVO size. Prenatal echocardiography should examine changes in CVOs and the occurrence of cardiovascular dysfunction. In general, the postpartum prognosis of fetal CVO is good.

Many corporations and nations have pledged to reach net-zero emissions within a few decades. Meeting such targets for greenhouse gases, plastics, etc. requires systematic methods to guide investment in technologies and value-chain alternatives, and develop roadmaps. The proposed framework is a multi-period planning model to guide optimal reforms in cradle-to-cradle life-cycle networks across the time horizon. It aims to meet environmental targets while minimizing the total annualized marginal cost of natural resources and the investment cost associated with adoption of novel technologies. This considers the evolution of technology readiness levels as S-curves or continuous time Markov-chains. Integrated Assessment models account for climate change, decarbonization due to energy mix changes, and carbon taxes. Multiple climate change scenarios and shared socioeconomic pathways are used to model the future. In addition to providing roadmaps, the outputs can also be used to identify technologies that will be robust to future scenarios.

Swelling granular media can experience size-induced percolation phenomena giving rise to segregation. In this work, the Discrete Element Method is employed to investigate the effects of size ratio and swelling kinetics on the segregation. The numerical analysis was carried out on a binary mixture consisting of coarse and fine particles and several mixing indices found in literature were adapted and tested for evaluating the mixing of expanding systems. Additionally, a relative percolation velocity was employed to quantify the percolation of fine particles. The results show that the percolation of fine particles becomes more significant as the size ratio increases. Additionally, results showed that the swelling kinetics has no impact on the segregation tendency. This research provides valuable insight into the effect of size ratio and swelling kinetics on the segregation behaviour of swelling granular materials, which can contribute to understanding percolation phenomena in various fields.

The term ‘habitat fragmentation’ is frequently associated with the biologically-destructive activities of human development, but an important evolutionary hypothesis posits that much of the biodiversity we see today was generated by episodic, natural habitat fragmentation. This hypothesis suggests that fragmentation can serve as a ‘crucible of evolution’ through the amplifying feedbacks of colonization, extinction, adaptation, and speciation. Interrogating the generality of this hypothesis requires measuring the repercussions of fragmentation at intra- and interspecific levels across entire communities. We use DNA metabarcoding to capture these repercussions from the scales of intraspecific differentiation to community composition in a megadiverse, ecologically foundational group, arthropods, using a natural habitat fragmentation experiment on patches of wet forest isolated by contemporary Hawaiian lava flows (kīpuka). We find a pronounced effect of area in kīpuka cores, where the taxonomic richness supported by a kīpuka scales with its size. Kīpuka cores exhibit higher intra- and interspecific turnover over space than continuous forest. Additionally, open lava, kīpuka edges, and the cores of small kīpuka (which are essentially entirely “edge”) host lower richness, are more biologically homogeneous, and have higher proportions of non-native taxa than kīpuka cores. Our work shows how habitat fragmentation isolates entire communities of habitat specialists, paving the way for genetic differentiation. Parsing the extent to which differentiation in kīpuka is driven by local adaptation versus drift provides a promising future avenue for understanding how fragmentation, and the different isolated communities created through this process, may lead to speciation in this system.

The capacity to form biofilms is a common trait among many microorganisms present on Earth. In this study, we demonstrate for the first time that the fatal pine pitch canker agent, Fusarium circinatum, can lead a biofilm-like lifestyle with aggregated hyphal bundles wrapped in extracellular matrix (ECM). Our study suggests that F. circinatum biofilms respond to a changing environment, demonstrated by poor and optimal biofilm development under particular abiotic conditions, including temperature and pH. Further analysis revealed that while planktonic cells produced small amounts of ECM per unit of the biomass, heat- and azole-exposed biofilms produced significantly more ECM than non-exposed biofilms. The increased synthesis of ECM in biofilms due to these abiotic factors underscores biofilm importance in response to various stress conditions, demonstrating the adaptability of F. circinatum to changing environments. Interestingly, azole exposure also led to biofilms that were resistant to DNase, which typically uncouples biofilms by penetrating the biofilm and degrading its extracellular DNA; we propose that DNases were likely hindered from reaching target cells by the ECM barricade. The interplay between antifungal treatment and DNase enzyme suggests a complex relationship between eDNA, ECM, and antifungal agents in F. circinatum biofilms. Therefore, our results show how a phytopathogen’s sessile (biofilm) lifestyle could influence its response to the surrounding environment.

The study of the diet of extreme specialist species can help to understand the selective pressures that modulate the trophic strategies of raptors. This article provides insight into the trophic strategy of a little-known top predator and allows for a better understanding of the selective pressures that shape its diet. We studied the diet of European honey-buzzards, a raptor specialized in the consumption of social wasp brood. The ratio of predator to prey biomasses is one of the highest among raptors. We studied some factors that affect the energy demand of pairs which influence the diet composition and the daily rate of prey delivery to the nest. We explore hypotheses about the role of native and exotic vespids in the diet and the influence of the number and age of nestlings on diet composition, daily rate of prey delivery, and size of combs delivered. We installed trail cameras in 24 honey-buzzards nests in north-western Spain in 2018-2021. We estimated the proportion and daily rate of delivery of native common-wasps (Vespula vulgaris), invasive Asian-hornets (Vespa velutina), reptiles, and birds and the honey-buzzard´s preferences for vespid species. We performed LMMs, GLMMs, and MLRMs to analyse relationships between response variables and predictors. We identified 4611 prey. Honey-buzzards mainly consumed vespids (82% of prey). Common-wasps and Asian- hornets were almost the only two vespids consumed. The invasive hornet was the second most important prey consumed, but common-wasps were preferred. Vespids were more important as the age and number of nestlings increased. Our results suggest that the honey-buzzard´s diet is the adaptive result of the conflict between being a median-sized insect-eating migratory nidicolous raptor and collecting enough morsels for the growth of its nestlings, supporting the prey size and ingestion rate hypothesis. Muscle-specific kinase myasthenia gravis (MuSK MG) is caused by autoantibodies against MuSK in the neuromuscular junction (NMJ). MuSK MG patients have fluctuating, fatigable weakness, in particular of bulbar muscles. Severity differs greatly between patients, in spite of comparable autoantibody levels. One explanation for inter-patient and inter-muscle variability in sensitivity might be variations in compensatory muscle responses. Previously, we developed a passive transfer mouse model for MuSK MG. In preliminary ex vivo experiments we observed that muscle contraction, in particular of mice with milder myasthenia, had become partially insensitive to μ-Conotoxin-GIIIB, a blocker of skeletal muscle NaV1.4 voltage-gated sodium channels. We hypothesized that changes in NaV channel expression profile, possibly co-expression of (μ-Conotoxin-GIIIB insensitive) NaV1.5 type channels, might lower the muscle fibre’s firing threshold and facilitate neuromuscular synaptic transmission. To test this, we here performed passive transfer in mice, using ‘high’, ‘intermediate’ and ‘low’ dosing regimens of purified MuSK MG patient IgG4 and compared myasthenia levels, μ-Conotoxin-GIIIB resistance, muscle fibre action potential characteristics and firing thresholds. High- and intermediate-dosed mice showed clear, progressive myasthenia, not seen in low-dosed animals. However, diaphragm NMJ electrophysiology demonstrated almost equal myasthenic severities amongst all regimens. Nonetheless, low-dosed mouse diaphragms showed a much higher degree of μ-Conotoxin-GIIIB resistance. This was not explained by upregulation of Scn5a (the NaV1.5 gene), lowered muscle fibre firing thresholds or histologically detectable upregulated NaV1.5 channels. It remains to be established which factors are responsible for the μ-Conotoxin-GIIIB insensitivity and whether the NaV repertoire change is compensatory beneficial, or a bystander effect.

Objective: Evaluate NHS-funded fertility treatment policies across England, Scotland, and Wales, assessing their alignment with the needs of those trying to conceive and comparing eligibility against NICE guidance. Design: Review of current NHS-funded fertility treatment policies and retrospective analysis of participant data. Setting: Not applicable. Population: Reproductive-age UK-based women using Hertility Health services from 2021-2023, focusing on those actively trying to conceive (TTC) or planning future pregnancies. Methods: Policy evaluation and data review without interventions. Main Outcome Measures: Eligibility for NHS-funded fertility treatment under UK regional policies versus NICE guidance. Results: In total, out of 2980 individuals actively trying to conceive and 6282 individuals planning for future pregnancies, 1700 (57.0%) and 2750 (43.8%) respectively would be ineligible for state-funded treatment under current regional policies: 1551 (57.7%) in England, 123 (53.2%) in Scotland, and 26 (41.9%) in Wales for those actively trying to conceive, and 2558 (43.9%), 146 (41.7%), and 46 (44.2%) for those planning future pregnancies respectively. In contrast, only 248 (8.3%) of those actively trying to conceive and 149 (2.4%) of those planning for future pregnancies would be ineligible if the NICE guidance were universally applied. Conclusions: By imposing outdated clinical and non-clinical qualifying criteria, the current NHS-funded fertility treatment policies exclude a substantial number of individuals requiring treatment. This necessitates a swift policy overhaul, as promised in the 2022 UK Women’s Health Strategy, to avoid further age-related fertility loss in women who require treatment imminently.

The current knowledge on insects preying on fruits is limited, and some of the scarce existing data on fruit-associated insects are secluded within the host institutions. Consequently, their value is not fully realized. However, the integration and interlinking of historical biocollections data of plants, fruits, and insects, collected in Kenya, within a digital framework have not been fully exploited. This necessitates the need to enhance accessibility by consolidating the historical biodiversity data onto a unified platform. To address these gaps, this article presents a description of the development of a web-based platform for data sharing and integrating biodiversity historical data of wild plants, fruits, associated insects, and their molecular barcodes (WiPtFruIM) while leveraging data science technologies. The platform holds invaluable potential in fruit pest management, by providing information on potential biocontrol agents for fruit pests, which can function as a decision-making tool and fruit-pest ecological modeling. The platform is invaluable information to a worldwide community (such as researchers, classroom education, nature enthusiasts, fruit pest management, modeling, etc.) to make informed decisions and build innovative tools. Rationale. Hydrogen/deuterium exchange mass spectrometry (HDX-MS) can provide precise analysis of a protein’s conformational dynamics across varied states, such as heat-denatured vs. native protein structures, localizing regions that are specifically affected by such conditional changes. Maximizing protein sequence coverage provides high confidence that regions of interest were located by HDX-MS, but one challenge for complete sequence coverage is N-glycosylation sites. The deuteration of peptides post-translationally modified by asparagine-bound glycans (glycopeptides) has not always been identified in previous reports of HDX-MS analyses, causing significant sequence coverage gaps in heavily glycosylated proteins and uncertainty in structural dynamics in many regions throughout a glycoprotein. Methods. We detected deuterated glycopeptides with a Tribrid Orbitrap Eclipse mass spectrometer performing data-dependent acquisition. An MS scan was used to identify precursor ions, if high-energy collision-induced dissociation (HCD) MS/MS of the precursor indicated oxonium ions diagnostic for complex glycans then electron transfer low-energy collision-induced dissociation (EThcD) MS/MS scans of the precursor identified the modified asparagine residue and the glycan’s mass. As in traditional HDX-MS the identified glycopeptides were then analyzed at the MS level in samples labeled with D 2O. Results. We report HDX-MS analysis of the SARS-CoV-2 spike protein ectodomain in its trimeric pre-fusion form, which has 22 predicted N-glycosylation sites per monomer, with and without heat treatment. We identified glycopeptides and calculated their average isotopic mass shifts from deuteration. Inclusion of the deuterated glycopeptides increased sequence coverage of spike ectodomain from 76% to 84%, demonstrated that glycopeptides had been deuterated, and improved confidence in results localizing structural re-arrangements. Conclusion. Inclusion of deuterated glycopeptides improves the analysis of the conformational dynamics of glycoproteins such as viral surface antigens and cellular receptors.
